Which desktop...
Like most Linux/Unix users I've been a big KDE fan for years. However, recently I've been running in Gnome full time. Gnome 2.6 has made some improvements that were show stoppers for me previously.

1) The "Open File" dialog was really poor in prior Gnome releases. 2.6's "Open File" dialog is similar to KDE and Windows. Actually it seems a little simpler -- I like it better.

2) The GTK+ library was a little slow for my taste. Gnome 2.6 uses GTK+ 2.4 and seems a bit snappier. This may be due in part to the 2.6 kernel as well, but the net effect is a faster Gnome.

Less important to me, but useful for beginners is a "My Computer" on the desktop. Although "My Computer" doesn't serve a huge purpose, many novice users who are migrating from Windows are used to it being there. Gnome 2.6 has a "Computer" icon on the desktop that serves this purpose.

KDE is great. I'll keep running both for a while, but I think that Miguel de Icaza has his head on straight as far as the design and future of Gnome. Mono (.NET for Linux/Unix), Java, Perl, Python, C/C++, and nearly all other popular programming languages can easily tie into Gnome. You can create commercial software for Gnome more easily that with KDE (KDE has issues with the Qt lib it's based on). All and all I see Gnome pulling ahead of KDE in features and commercial/OSS supporters in the near future. Gnome 2.6 is clean and easy -- what more could you want?

At the end of the day, both Gnome and KDE are superb in there current state. You can't loose.
