Two BIG things wrong
Two major flaws:

1) Mail could only be delivered when the sender is online. I, for one, occasionally send mail from my laptop and that isn't always on the internet. This idea would require me to either stop sending mail from computers that weren't alway-on, or give up on quick delivery.

2) This would require every user who uses e-mail to open an inward facing, Internet connected port open on their computers. You think spam's a problem? Imagine the security issues with what is effectively an easily accessable, open file-sharing port on every computer on the planet.
